  Capability extensions that unblock GenoADME deferred Tier 1/2 pairs (NAT2/isoniazid, UGT1A1/irinotecan, SLCO1B1/simvastatin, CYP2C19/clopidogrel). Issues are independently actionable; the milestone closes when GenoADME can promote at least one Deferred pair to an active tier and produce a passing Tier 1/2 result.

• v0.2: Tier 1 PASS (calibration)

  Calibration work that closes the GenoADME Tier 1 PARTIAL gap. After the 2026-05-01 reproducibility audit (GenoADME commit 349110b), the gap is the PM/EM AUC ratio over-shoot (2.737 vs [1.4, 2.5] band), not AAFE (which now PASSES). Sub-issues: #12 (#8a, XGBoost↔ECM reconciliation — surfaced the model-level mismatch), #13 (#8b, test_oatp_pravastatin invariant), #14 (#8c, abundance re-calibration). Note: empirical PoC of #8a alone showed it cannot close the PM/EM ratio band (over-shoots to ~4.4 from baseline 2.7); option (A1) explicit non-OATP CL term per drug is the most likely path. Umbrella issue #8 closes when GenoADME re-run produces all three criteria PASS via a metric-change: commit.
