Hello everyone, we are now at the twelfth week of Cytometry in R course! For previous course emails, see here.
This week for the primary course material, we will explore how to extract fluorescent signatures from our unmixing controls (both single-color and unstained). In the process, we deep dive into what often seems like a black box when we first start to learn about spectral flow cytometry, with the goal of demystifying what goes on behind the scenes, allowing us to better decide what makes a good quality unmixing control for our own experiments. Additionally, we will highlight some of the new functions that are present in both the flowGate and Luciernaga packages that make this workflow far easier than it has been previously to implement in R. You can find this week's course material here.
For those in Baltimore attending in-person:
For those attending online, our YouTube live-streams will be unfortunately a little off-schedule this week. For our primary course material, the live-streams will be this week at:
Wednesday 2200 EST (Thursday 0200 GMT+0)
Thursday 1000 EST (Thursday 1400 GMT+0)
Friday 1600 EST (Friday 2000 GMT+0)
All recordings will be available immediately afterwards on YouTube.
Additionally, next week I will try to record all the one-off bonus walk-throughs (since I have commitments on Wednesday and Thursday). These are currently set for:
Monday 1800 EST (Monday 2200 GMT+0), covering the Concatenate Extended; Retrieving ImmPort datasets; and Clean Up Gate bonus materials.
Tuesday 1800 EST (Tuesday 2200 GMT+0), covering Signature Matrices; and LuciernagaIntegration.
Potentially Friday same time for AutoSpectral and TRU-OLS recording, will confirm for next weeks email.
Best Wishes-
David
